CDKN1A (Cyclin-Dependent Kinase Inhibitor 1A), also known as p21, is a crucial regulator of the cell cycle. It inhibits cyclin-dependent kinases (CDKs) to halt cell cycle progression at the G1 phase, primarily in response to DNA damage. p21 is transcriptionally activated by p53 and plays a key role in maintaining genomic stability by facilitating DNA repair. It also modulates apoptosis and cellular senescence. Loss of CDKN1A function is linked to uncontrolled proliferation in cancer, while its overexpression can contribute to therapy resistance.
