Introduction:

The Dynamin 1 like protein is the product of the DNM1L gene. This disease related protein serves an important role by mediating membrane fission, ensuring the survival of several types of postmitotic neurons and controlling mitochondrial ATP production. The DNM1L protein can be found in several human tissues and mutations or disfunctions of its gene lead to mental disorders, cancers and lethal encephalopathy. What is DNM1L?

The protein encoded by the DNM1L gene functions in mitochondrial and peroxisomal division and mediates membrane fission through oligomerization into membrane-associated tubular structures that wrap around the scission site to constrict and sever the mitochondrial membrane through a GTP hydrolysis-dependent mechanism. While the recruitment by the membrane receptors is GTP-dependent, the following hydrolysis of GTP induces the dissociation from the receptors and allows DNM1L filaments to curl into closed rings that are probably sufficient to sever a double membrane. Through its function in mitochondrial division, the DNM1L protein ensures the survival of at least some types of postmitotic neurons, including Purkinje cells, by suppressing oxidative damage and it is required for normal brain development, including that of cerebellum. This protein is required for programmed necrosis execution and the rhythmic control of its activity following phosphorylation at Ser-637 is essential for the circadian control of mitochondrial ATP production.

Where is DNM1L found?

Inside the cell, DNM1L is primairly found in the cytosol and additionally in the vesicles, and it has granular cytoplasmic and membranous expression in several tissues.

Dysfunction of this gene is implicated in several neurological disorders, including Alzheimer's disease and mutations in this gene are associated with the autosomal dominant disorder, encephalopathy, lethal, due to defective mitochondrial and peroxisomal fissionĀ
