The DDDDK-Tag is a compact epitope tag incorporating an enterokinase cleavage site in itself that allows for the complete removal of the tag from the fused proteins.
Although over the years numerous attempts have been made to optimize the FLAG-sequence via ELISA or phage display, the original sequence DYKDDDDK is still used for virtually all FLAG-tagged proteins.
