Post Translational Modifications Phosphorylation at Ser-348 by SGK1 inhibits its activity. Dephosphorylation of Ser-348 by the SHOC2-MRAS-PP1c (SMP) complex consisting of SHOC2, GTP-bound M-Ras/MRAS and the catalytic subunit of protein phosphatase 1 (PPP1CA, PPP1CB or PPP1CC).this relieves inactivation and stimulates kinase activity. Methylation by PRMT5 decreases stability and kinase activity. Ubiquitinated by RNF149.which leads to proteasomal degradation. Polyubiquitinated at Lys-615 in response to EGF.
Function Involved in the transduction of mitogenic signals from the cell membrane to the nucleus. Phosphorylates MAP2K1, and thereby activates the MAP kinase signal transduction pathway. Phosphorylates PFKFB2. May play a role in the postsynaptic responses of hippocampal neurons.
