| Post Translational Modifications | NMDA is probably regulated by C-terminal phosphorylation of an isoform of GRIN1 by PKC. Dephosphorylated on Ser-897 probably by protein phosphatase 2A (PPP2CB). Its phosphorylated state is influenced by the formation of the NMDAR-PPP2CB complex and the NMDAR channel activity. |
| Function | Component of N-methyl-D-aspartate (NMDA) receptors (NMDARs) that function as heterotetrameric, ligand-gated cation channels with high calcium permeability and voltage-dependent block by Mg(2+). NMDARs participate in synaptic plasticity for learning and memory formation by contributing to the long-term potentiation (LTP). Channel activation requires binding of the neurotransmitter L-glutamate to the GluN2 subunit, glycine or D-serine binding to the GluN1 subunit, plus membrane depolarization to eliminate channel inhibition by Mg(2+). NMDARs mediate simultaneously the potasium efflux and the influx of calcium and sodium. Each GluN2 or GluN3 subunit confers differential attributes to channel properties, including activation, deactivation and desensitization kinetics, pH sensitivity, Ca2(+) permeability, and binding to allosteric modulators. |

Frequently Asked Questions
Common questions about Mouse NMDAR1 (NMDAR1) Antibody Pair (STJAP00803)
This antibody pair is designed to detect mouse NMDAR1 (N-Methyl-D-Aspartate Receptor Subunit 1), corresponding to UniProt accession P35438. Reactivity has been established for mouse samples only.
This antibody pair is validated for use in ELISA. It is intended for sandwich ELISA configurations using the two provided monoclonal antibodies as capture and detection reagents.
Both antibodies are supplied as unconjugated, affinity-purified rabbit monoclonal IgGs at 1 mg/mL, formulated in 0.01M PBS at pH 7.2. Clone IDs are R14-6Y-3 and R84-4W-8 for the two components.
Working concentrations are not pre-specified and must be determined by the end user through titration optimization for their specific experimental conditions.
Both antibodies should be stored at 4°C for short-term use. For long-term storage, aliquot and store at -20°C, and avoid repeated freeze/thaw cycles to maintain activity.
