Additional Info
| Tissue Specificity | Widely expressed with highest levels in lymphoid tissues. Detected in activated T-cells where expression levels are 30- to 50-fold less than CD28, the stimulatory coreceptor, on the cell surface following activation. |
| Post Translational Modifications | N-glycosylation is important for dimerization. Phosphorylation at Tyr-201 prevents binding to the AP-2 adapter complex, blocks endocytosis, and leads to retention of CTLA4 on the cell surface. |
| Function | Inhibitory receptor acting as a major negative regulator of T-cell responses. The affinity of CTLA4 for its natural B7 family ligands, CD80 and CD86, is considerably stronger than the affinity of their cognate stimulatory coreceptor CD28. |

Frequently Asked Questions
Common questions about Human CTLA4 (CTLA4) Antibody Pair (STJAP00647)
This antibody pair detects human CTLA-4 (UniProt P16410). Reactivity has been confirmed for human samples only.
Both antibodies are supplied as affinity-purified monoclonal IgGs at 1 mg/mL in liquid format. The capture antibody (clone 1A8, IgG1) is formulated in PBS with 0.09% sodium azide, and the detection antibody (clone 9C7, IgG2a) is formulated in PBS with 0.01% thimerosal. Neither antibody carries a conjugation or tag.
This antibody pair has been validated for ELISA only, at a dilution of 1:1000.
Both antibodies were raised in mouse and are monoclonal. Clone 1A8 is an IgG1 isotype and clone 9C7 is an IgG2a isotype, both generated against a synthesized peptide derived from human CTLA-4.
Both antibodies should be stored at 4°C for short-term use. For long-term storage, aliquot and store at -20°C. Repeated freeze/thaw cycles should be avoided to maintain antibody integrity.
