Introduction:

DAB adaptor protein 1 is a protein synthesised from the DAB1 gene and is responsible for normal brain development and is thought to be a signal transducer interacting with protein kinase pathways. Additionally, DAB1 can mainly be found in the cerebellum and MAIT T-cells and mutations of it can lead to several forms of ataxia and the Marinesco-Sjgren Syndrome. What is DAB1?

DAB1 is a protein coding gene. The laminar organization of multiple neuronal types in the cerebral cortex is required for normal cognitive function. For example in mice, the disabled-1 gene plays a central role in brain development, directing the migration of cortical neurons past previously formed neurons to reach their proper layer. The DAB1 gene is similar to disabled-1, and the protein encoded by this gene is thought to be a signal transducer that interacts with protein kinase pathways to regulate neuronal positioning in the developing brain.

Where is DAB1 found?

DAB1 is usually found in the vesicles of the cells and is mainly found in the brain and intestine on tissue level. Inside the brain, it is enriched in the cerebellum and in the immune system it is expressed mainly in the MAIT T-cells.

What role does DAB1 play in disease?

Mutations of this gene are related to spinocerebellar ataxia 37, presenting progressive incoordination of the limbs, eyes and speech due to degeneration of the cerebellum.
