Introduction:

Caspase 8 is the protein produced by the CASP8 gene and is responsible for the execution phase of programmed cell death and its cleaving to another enzyme is crucial for limiting apoptosis and necroptosis during embryonic development. It can be used as a prognostic marker in unfavourable renal and liver cancer and favourable ovarian cancer. What is CASP8?

This gene encodes a member of the cysteine-aspartic acid protease (caspase) family and the sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. The synthesised protein is the most upstream protease of the activation cascade of caspases responsible for the TNFRSF6/FAS mediated and TNFRSF1A induced cell death. Binding to the adapter molecule FADD recruits it to either receptor and the resulting aggregate called death-inducing signaling complex (DISC) performs CASP8 proteolytic activation. The active dimeric enzyme is then liberated from the DISC and free to activate downstream apoptotic proteases. It cleaves and activates CASP3, CASP4, CASP6, CASP7, CASP9 and CASP10 and also it may be a target for the cowpox virus CRMA death inhibitory protein. CASP8 cleaves RIPK1 at 'Asp-325' which is crucial for limiting apoptosis and necroptosis during embryonic development.

Where is CASP8 found?

CASP8 can be found in various places inside the cell, most dominantly in the cytosol and nucleoplasm and its protein is expressed with varying intensity in most tissues.

What role does CASP8 play in disease?

This protein is expressed in the insoluble fraction of the affected brain region from Huntington disease patients but not in those from normal controls, which implicated its role in neurodegenerative diseases. Apart from that, an ALPS- resembling disorder named Caspase 8 deficiency is triggered by a homozygous mutation in the CASP8 gene and other abnormalities in the gene and its expression lead to breast and lung cancer.
