Introduction:

The CASP1 gene encodes the information for synthesising the Caspase 1 protein, responsible for both defence from pathogens and several inflammatory processes, but apart from its useful roles in the human body, mutations or overexpressions can lead to conditions such as Familial Mediterranean Fever, cowpox, different types of cancers and shingellosis. What is CASP1?

This gene encodes a protein which is a member of the cysteine-aspartic acid protease (caspase) family and is responsible for sequential activation of caspases and plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce 2 subunits, large and small, that dimerize to form the active enzyme. The product of this gene is a thiol protease that cleaves IL-1 beta between an Asp and an Ala, releasing the mature cytokine which is involved in a variety of inflammatory processes also, it is important for defense against pathogens. Another function if the protein is to cleave and activate sterol regulatory element binding proteins (SREBPs).

Where is CASP1 found?

Inside the cell, the protein can be found both in the cytosol and the nucleoplasm. Apart from that, it can be found in the majority of human tissues, but it is enhanced in the blood and lymphoid tissue.

What role does CASP1 play in disease?

Being a cancer related gene, the protein that it encodes can be found in majority of cancers. Furthermore, it the gene is associated with cowpox because some of the pathways are Innate Immune System(IIS) and Apoptotic Pathways in Synovial Fibroblasts(APSF). The ISS pathway of this gene is also related with shingellosis, and mutations are responsible for an autoinflammatory disorder called Familial Mediterranean Fever.
