Introduction:

Baculoviral IAP repeat containing 5 is a gene that encodes a protein responsible for cell proliferation and preventing apoptosis and participating in the chromosome passage protein complex. What is BIRC5?

This gene is a member of the inhibitor of apoptosis (IAP) gene family, which encode negative regulatory proteins that prevent apoptotic cell death and usually contain multiple baculovirus IAP repeat (BIR) domains, but this gene encodes proteins with only a single BIR domain.It encodes a multitasking protein that has dual roles in promoting cell proliferation and preventing apoptosis, and also is a component of a chromosome passage protein complex (CPC) which is essential for chromosome alignment and segregation during mitosis and cytokinesis. The protein acts as an important regulator of the localization of this complex and directs CPC movement to different locations from the inner centromere during prometaphase to midbody during cytokinesis and participates in the organization of the center spindle by associating with polymerized microtubules. It is involved in the recruitment of CPC to centromeres during early mitosis via association with histone H3 phosphorylated at 'Thr-3' (H3pT3) during mitosis.

Where is BIRC5 found?

BIRC5 is a protein which is normally expressed in embryonic tissues and during fetal development, as well as in fast dividing normal cells (like bone marrow stem cells, basal epithelial cells and thymocytes, even if at lower concentrations), but is virtually undetectable in fully differentiated and healthy adult tissues.  

What role does BIRC5 play in disease?

An elevated expression of BIRC5 protein has been found in almost all brain, breast, colorectal and gastric cancer. Estrogen positive breast cancer subtypes have been connected to increased BIRC5 regulation, which was reverted by treatment with the natural compound myricetin, enhancing apoptosis.
