Introduction:

Aryl hydrocarbon receptor nuclear translocator(ARNT) gene provides the information for synthesising the ARNT protein, which is responsible for the normal functioning of the hypoxia response element and also functions as a transcriptional regulator of the adaptive response to hypoxia. What is ARNT?

The protein encoded by this gene is required for activity of the Ah (dioxin) receptor and for the ligand-binding subunit to translocate from the cytosol to the nucleus after ligand binding. The complex then initiates transcription of genes involved in the activation of PAH procarcinogens. The heterodimer binds to core DNA sequence 5'-TACGTG-3' within the hypoxia response element (HRE) of target gene promoters and functions as a transcriptional regulator of the adaptive response to hypoxia. The heterodimer ARNT:AHR binds to core DNA sequence 5'-TGCGTG-3' within the dioxin response element (DRE) of target gene promoters and activates their transcription.

Where is ARNT found?

On intracellular level, the ARNT protein can be found in the nucleoplasm and the nuclear bodies. Inside the human organism, both the gene and the expressed protein have low tissue specificity and can be detected in all tissues.

What role does ARNT play in disease?

Because it is connected with the HRE, disruptions in the protein or gene can lead to bad responses to environmental stresses and inducing general or tissue hypoxia. Apart from that, several mutations have been found in human cancers, with low cancer specifity.
