Detection of polyhistidine-tagged fusion proteins purified by immobilised metal affinity chromatographyThe polyhistidine tag, or His-Tag, is the most widely used affinity tag for recombinant protein purification. It consists of 6 to 10 consecutive histidine residues at one terminus of the target protein, often separated from it by a protease-cleavage site, and binds selectively to immobilised metal ions such as nickel in a technique first described by Hochuli and colleagues in 1988.1
Because the tag is so small, it is usually left on the recombinant protein without needing to be removed, since it has little to no effect on the protein's structure or activity. Our range covers mouse monoclonal and rabbit polyclonal formats across Western Blot, IF/ICC, IP and ELISA.
