Detection of FLAG-tagged fusion proteins carrying the eight-residue DYKDDDDK peptideThe FLAG octapeptide (DYKDDDDK) is a short, hydrophilic marker sequence engineered onto the N-terminus of recombinant proteins to aid their detection and purification, using an antibody raised against the first four residues of the tag.1 Because the peptide is small and unrelated to the biology of most fusion partners, tagged proteins generally retain their biological activity.
The tag can be added to either the N- or C-terminus of a protein, or used in tandem repeats, and its mild elution conditions make it suitable for isolating whole multi-subunit protein complexes rather than just the tagged protein alone.
