• Western blot of GFP-Cas9 transfected HEK293 cell lysate (+) showing specific immunolabeling of the ~53 kDa GFP-Cas9 fusion protein. Labeling is absent in the untransfected lysate (-).
  • Immunofluorescence of HEK293 cells transfected with a fusion construct of GFP with the C-terminal region of S. aureas Cas9 protein (green) that is labeled with Anti-CRISPR Cas9 (304-Cas9, 1:1000, red) and the DNA stained blue with DAPI. The yellow label is the overlapping of the Anti-Cas9 antibody and the Cas9-GFP construct. m

Background CRISPR/Cas9 is a genome editing tool that makes possible the permanent alteration of genes by directing specifically designed RNA sequences complexed with the nuclease Cas9 to cleave DNA at any desired location allowing the deletion or addition of genes. Antibodies against Cas9 are very useful in the direct detection of the CRISPR/Cas9 complex. This antibody detects Cas9 from Staphylococcus aureus transfected into cells and shows no reactivity toward the Streptococcus pyogenes Cas9 protein (Ran et al., 2015).
