A standard whole-IgG (H+L) secondary detects both the heavy and light chains of a primary antibody, and only works if the primary carries a conventional Fc region. The two products below cover cases outside that: a secondary raised against a nanobody rather than an Fc region, and a secondary that reads the light chain only.
Rabbit Anti-Llama VHH Antibody {HRP} (STJS000855)
Camelid nanobodies (VHH domains) have no conventional IgG Fc region, so a standard anti-Fc secondary will not detect them. This secondary is raised directly against the VHH domain, validated by western blot at 1:500-1:1000. An unconjugated format is also available.

Goat Anti-Mouse IgG Light Chain Antibody {HRP} (STJS001198)
Binds only the light chain of mouse IgG, not the heavy chain. Useful when a co-immunoprecipitation or hybridoma western blot would otherwise pick up heavy-chain signal from the IP antibody itself. Validated at 1:5000 dilution against mouse IgG.

Secondary Antibodies for Flow Cytometry and ICC/IF
Fluorophore-conjugated secondaries are used for cell-surface and intracellular staining in flow cytometry, and for direct visualisation of target localisation by immunofluorescence and immunocytochemistry. The Alexa Fluor™ products below are all validated for fluorescence microscopy and flow cytometry.

Choosing, Diluting and Troubleshooting Your Secondary Antibody
Selecting the right secondary comes down to three variables: target species, isotype and conjugation, matched against the primary antibody and detection system in use.
Why use secondary antibodies?
Signal amplification: multiple secondary antibodies can bind a single primary antibody, increasing detection sensitivity, especially for low-abundance targets. Versatility: compatible with a wide range of detection systems and formats. Multiplexing: enables simultaneous detection of multiple targets using different fluorophores.
Choosing the right secondary antibody
Target species should match the primary antibody host species. Isotype should match the primary antibody isotype. Conjugation should be chosen according to application needs. All secondary antibodies in the range are affinity-purified, giving high specificity and low background.
Optimising dilution
Primary antibody: typical dilution 1:100 to 1:1,000. Secondary antibody: recommended dilution 1:500 to 1:2,000 for immunofluorescence. Excessive concentration can lead to high background or non-specific staining.
Troubleshooting tips
Use cross-adsorbed secondary antibodies, which have additional purification steps to remove off-target immunoglobulin, reducing cross-reactivity. Include negative controls (no primary antibody) to assess background. Adjust blocking and washing steps to minimise immunofluorescence background.
