Photostable, pH-stable fluorescent dyes for flow cytometry, imaging and multiplex detection
Alexa Fluor dyes are conjugated directly to the secondary antibody. Once bound to a primary antibody, the dye is excited at a set wavelength and emits a photon at a longer wavelength, which is captured by a fluorescence microscope or flow cytometer as the detection signal. Alexa Fluor dyes are more photostable and pH-stable than the fluorophores they were synthesised from, and their low species cross-reactivity supports multi-colour panels where several targets are labelled and read at once. Our range covers Alexa Fluor conjugates against goat, donkey, mouse, rabbit and other host and reactivity combinations, spanning wavelengths from 350 nm to 790 nm.

A goat anti-mouse IgG (H&L) Alexa Fluor 488 conjugate is the standard green-channel secondary for detecting mouse primaries by immunofluorescence and flow cytometry. A goat anti-rabbit IgG (H&L) Alexa Fluor 647 conjugate, highly cross-adsorbed against mouse, rat, bovine, goat and human IgG, pairs with it in the same multi-colour panel without one secondary picking up the other species' primary.
Goat Anti-Mouse IgG (H&L) Antibody {Alexa Fluor 488}
An affinity-purified goat polyclonal secondary conjugated to Alexa Fluor 488, for detecting mouse-raised primaries in fluorescence microscopy and flow cytometry. Recommended dilutions are 1 to 2 µg/mL for fluorescence microscopy and 1 µg per 10⁶ cells for flow cytometry.

Mouse and rabbit are the most common host species for monoclonal and polyclonal primaries, so goat anti-mouse and goat anti-rabbit secondaries make up the core of the range. Both reactivities are available as standard IgG (H&L) conjugates or as highly cross-adsorbed conjugates, which have been affinity purified against the IgG of other common host species to reduce cross-reactivity when mouse- and rabbit-raised primaries are run in the same panel.

Donkey Host Alexa Fluor Secondary Antibodies
Donkey-raised secondaries are commonly selected for multi-colour panels because donkey antisera are cross-adsorbed against the immunoglobulins of other host species used in the same panel, reducing the chance that one secondary picks up a primary it was not meant to detect. Our donkey Alexa Fluor secondaries cover anti-goat, anti-mouse, anti-rabbit and anti-rat reactivities, with the mouse, rabbit and rat conjugates highly cross-adsorbed for use alongside each other in the same experiment.

Anti-Chicken IgY Alexa Fluor Secondary Antibodies
Chicken IgY primaries are used where a mammalian-raised primary would cross-react with endogenous mammalian immunoglobulin in the sample, since IgY does not bind mammalian Fc receptors or complement in the same way as IgG. Our goat anti-chicken IgY Alexa Fluor conjugates cover the widest wavelength range in the line, from Alexa Fluor 350 through to Alexa Fluor 790, including the 680 and 790 conjugates used with laser- and filter-based infrared imaging instruments operating in the 700 nm and 800 nm channels.

Anti-Human Fragment-Specific Alexa Fluor Secondary Antibodies
Fab fragment and F(ab')2 secondaries detect human IgG through the antigen-binding region rather than the Fc region, which avoids binding to Fc receptors expressed on immune cells in the sample. This makes fragment-specific conjugates useful where an intact IgG secondary would generate Fc-mediated background, such as when staining immune cell populations for flow cytometry or detecting human antibodies in patient serum. Our goat anti-human range covers both Fab fragment and F(ab')2 formats across the Alexa Fluor wavelength range.

Matching a conjugate to your imaging instrument
Use a blue laser diode line for Alexa Fluor 405, a cyan 488 nm laser for Alexa Fluor 488, a yellow 526 nm laser for Alexa Fluor 550 or 594, and a red 633 nm laser for Alexa Fluor 649. Alexa Fluor 680 and 790 conjugates are for laser- and filter-based infrared imaging instruments emitting at 700 nm and 800 nm.
