Cyclin dependent kinase inhibitor 1A is a protein synthesised by the CDKN1A gene and is tightly controlled by the p53. What is CDKN1A?
This gene encodes a potent cyclin-dependent kinase inhibitor. The encoded protein binds to and inhibits the activity of cyclin-cyclin-dependent kinase2 or -cyclin-dependent kinase4 complexes, and thus functions as a regulator of cell cycle progression at G1. The expression of this gene is tightly controlled by the tumor suppressor protein p53, through which this protein mediates the p53-dependent cell cycle G1 phase arrest in response to a variety of stress stimuli. Also, this protein can interact with proliferating cell nuclear antigen, a DNA polymerase accessory factor, and plays a regulatory role in S phase DNA replication and DNA damage repair. Cyclin dependent kinase inhibitor 1A was reported to be specifically cleaved by CASP3-like caspases, which thus leads to a dramatic activation of cyclin-dependent kinase2, and may be instrumental in the execution of apoptosis following caspase activation.
Where is CDKN1A found?
CDKN1A is usually found in the nucleoplasm and nuclear bodies and despite having low tissue and brain region specificity it is immune cell enhanced.
What role does CDKN1A play in disease?
Being a cancer-related gene, CDKN1A is responsible for tongue and other forms of cancers. Apart from that, its relation to P53 leads to papilloma if abnormalities occur and bladder transitional cell papilloma if mutations of TP53 occur. Furthermore, if a mutation of the TP53 happens, Li-Fraumeni syndrome can occur.
