Introduction:
The protein encoded by the AXL gene is a member of the TAM receptor tyrosine kinase subfamily and may be responsible for several important cellular functions such as survival, migration and differentiation. In diagnostics it is used as a prognostic marker for unfavourable renal cancer and several mutations of AXL are linked with breast, thyroid and other human cancers. Furthermore, overexpressing of AXL and several other members of the TAM family result in a condition known as LCMV. What is AXL?
The protein encoded by this gene is a member of the Tyro3-Axl-Mer (TAM) receptor tyrosine kinase subfamily, and it possesses an extracellular domain, composed of two immunoglobulin-like motifs at the N-terminal, followed by two fibronectin type-III motifs. It transduces signals from the extracellular matrix into the cytoplasm by binding to the vitamin K-dependent protein growth arrest-specific 6 (Gas6), which is regulating many physiological processes including cell survival, cell proliferation, migration and differentiation. . This gene may be involved in several cellular functions including growth, migration, aggregation and anti-inflammation in multiple cell types. Alternative splicing results in multiple transcript variants of this gene.
Where is AXL found?
Inside the cell, AXL protein is found mainly in the plasma membrane, actin filaments and vesicles. Generally, it has low tissue specificity, but higher levels are found in male-specific tissues.
What role does AXL play in disease?
Several mutations have been reported in breast, thyroid cancer and adenocarcinoma. Apart from that, overexpression of AXL and other members of the TAM family enhance the lymphocytic choriomeningitis(LCMV) infecting rate of cells.
