Additional Info
| Function | Major immune regulatory cytokine that acts on many cells of the immune system where it has profound anti-inflammatory functions, limiting excessive tissue disruption caused by inflammation. Mechanistically, IL10 binds to its heterotetrameric receptor comprising IL10RA and IL10RB leading to JAK1 and STAT2-mediated phosphorylation of STAT3. In turn, STAT3 translocates to the nucleus where it drives expression of anti-inflammatory mediators. Targets antigen-presenting cells (APCs) such as macrophages and monocytes and inhibits their release of pro-inflammatory cytokines including granulocyte-macrophage colony-stimulating factor /GM-CSF, granulocyte colony-stimulating factor/G-CSF, IL-1 alpha, IL-1 beta, IL-6, IL-8 and TNF-alpha. Also interferes with antigen presentation by reducing the expression of MHC-class II and co-stimulatory molecules, thereby inhibiting their ability to induce T cell activation. In addition, controls the inflammatory response of macrophages by reprogramming essential metabolic pathways including mTOR signaling. |

Frequently Asked Questions
Common questions about Human IL-10 (Interleukin-10) Antibody Pair (STJAP00037)
This antibody pair is designed to detect human IL-10 (Interleukin-10, UniProt P22301) and has confirmed reactivity with human samples only. It is not validated for use with other species.
The capture antibody is a mouse monoclonal antibody supplied unconjugated, while the detection antibody is a goat polyclonal antibody supplied with a biotin conjugate. Both components are provided at 0.5 mg/mL in PBS with 0.04% Proclin 300 and 50% glycerol at pH 7.4.
This pair is validated for use in ELISA only. No other assay formats are listed as validated applications for this product.
The capture antibody was purified by Protein A or G, and the detection antibody was purified by antigen affinity purification. Both antibodies were raised against recombinant human IL-10 protein as the immunogen.
Both components should be stored at -20 degrees Celsius for long-term storage. Freeze-thaw cycles should be avoided to maintain antibody integrity.
