Introduction:

Discs large MAGUK scaffold protein 4 is the protein encoded by the DLG4 gene and takes part in the NMDA receptors and combined with another protein from the same family can interact at postsynaptic sites. The DLG4 protein can primairly be found in the brain and some cells of the immune system, but disorders of its expression or mutations can cause degeneration of the brain, several mental disorders and Huntington's disease. What is DLG4?

This gene encodes a member of the membrane-associated guanylate kinase (MAGUK) family.The potein which is sythesised heteromultimerizes with another MAGUK protein, DLG2, and is recruited into NMDA receptor and potassium channel clusters. These two MAGUK proteins may interact at postsynaptic sites to form a multimeric scaffold for the clustering of receptors, ion channels, and associated signaling proteins.

Where is DLG4 found?

This protein is expressed mainly in the brain tissue and several immune cells, including plasmacytoid DC and memory CD8 T-cell. Inside the cell, it is localised in the plasma membrane and cytoskeleton in higher quantities.

What role does DLG4 play in disease?

DLG4 is an important gene for cerebral degeneration. Apart from that, mutations of this gene can cause Huntington's disease, Schizophrenia and Autism Spectrum disorder. Furthermore, overexpression or depletion of DLG4 changes the ratio of excitatory to inhibitory synapses in hippocampal neurons.
