Introduction:

DNA damage inducible transcript 3 is a protein encoded by the DDIT3 gene. This protein plays the role of dominant-negative inhibitor and preventing DNA binding activity, but it also is implicated in adipogenesis and erythropoiesis which are activated by ER stress. Furthermore, it regulates inflammatory response trough caspase-11 induction and has cytoplasmic expression in most of the tissues. DDIT3 ca be found in the neutrophils and fusions with other genes can lead to several types of sarcomas. Apart from that, mutations of this gene can cause several types of cancers, fatty liver disease and skin disorders, but it can also be used as a prognostic marker in unfavourable liver cancer. What is DDIT3?

This gene encodes a member of the CCAAT/enhancer-binding protein (C/EBP) family of transcription factors. The DDIT3 protein functions as a dominant-negative inhibitor by forming heterodimers with other C/EBP members, such as C/EBP and LAP (liver activator protein), and preventing their DNA binding activity. This protein is implicated in adipogenesis and erythropoiesis, is activated by endoplasmic reticulum stress, and promotes apoptosis. Furthermore, it plays a regulatory role in the inflammatory response through the induction of caspase-11 (CASP4/CASP11) which induces the activation of caspase-1 (CASP1) and both these caspases increase the activation of pro-IL1B to mature IL1B which is involved in the inflammatory response.

Where is DDIT3 found?

DDIT3 can mainly be located in the nucleoplasm and it has nuclear and cytoplasmic expression in most tissues. In the immune system, DDIT3 can be found in neutrophils.

What role does DDIT3 play in disease?

Fusion of this gene and FUS on chromosome 16 or EWSR1 on chromosome 22 induced by translocation generates chimeric proteins in myxoid liposarcomas or Ewing sarcoma. Furthermore, mutations can cause fatty liver disease and several skin disorders.
