What is His-Tag
The polyhistidine tag, also known as His-Tag is the most common affinity tag used for the purification of recombinant proteins. It consists of 6 to 10 consecutive histidines located at one of the termini of the target protein and is frequently separated by a protease-cleavage site.
His-Tag structure
Because of its small size (6-10 amino acids), the His-Tag is usually left on the recombinant protein and doesn't need to be removed, as it has little to no effect on the structure of the protein.
