Background Zeta s Colon Cocktail – DUAL STAIN. Zeta s Colon Cocktail provides dual staining for Cadherin-17 (CDH17) and CDX-2, a valuable immunohistochemistry (IHC) technique primarily used in gastrointestinal (GI) pathology, especially to (1) identify tumors of intestinal origin, (2) differentiate between colorectal adenocarcinoma and other carcinomas, and (3) aid in metastatic tumor origin determination. Zeta s recombinant antibodies recognize Cadherin 17 and CDX-2, novel diagnostic markers for adenocarcinoma of the digestive system. Cadherin 17 (CDH17) is a novel oncogene expressed in intestinal epithelium which is involved in tumor invasion and metastasis. CDH17 is a highly specific marker in colon cancer. CDX2 is a homeobox gene that encodes an intestine-specific transcription factor. Anti-CDX2 has been useful to establish GI origin of metastatic adenocarcinomas and carcinoids and is especially useful to distinguish metastatic colorectal adenocarcinoma from lung adenocarcinoma. Colorectal carcinoma is one of most common cancers in human and it often metastasizes to liver or other locations before diagnosis. The combination AP-labeled rabbit (cadherin 17) (red) and HRP-labeled mouse (CDX-2) (brown) paraffin immunohistochemistry is highly sensitive and somewhat specific for colorectal and stomach adenocarcinoma. Zeta s Dual Stain Colon Cocktail provides increased diagnostic specificity for colorectal adenocarcinoma as both markers are highly expressed in colorectal cancers and dual staining ensures redundancy and confirmation, which can be helpful when one marker is weakly expressed or patchy. Additionally, the Colon Cocktail can help differentiate GI from non-GI tumors because CDX-2 can occasionally be expressed in non-GI tumors (e.g., mucinous ovarian carcinoma, bladder adenocarcinoma) but CDH17, being more restricted to intestinal epithelium, helps confirm true intestinal origin when paired with CDX-2. Finally, the Colon Cocktail can be useful in tumor origin identification in metastasis to evaluate liver, peritoneal, or lymph node metastases when the primary site is unknown. Tumors with CDH17⁺/CDX-2⁺ profile suggest a colorectal or small intestinal primary.
