Additional Info
| Function | Major immune regulatory cytokine that acts on many cells of the immune system where it has profound anti-inflammatory functions, limiting excessive tissue disruption caused by inflammation. Mechanistically, IL10 binds to its heterotetrameric receptor comprising IL10RA and IL10RB leading to JAK1 and STAT2-mediated phosphorylation of STAT3. In turn, STAT3 translocates to the nucleus where it drives expression of anti-inflammatory mediators. Targets antigen-presenting cells (APCs) such as macrophages and monocytes and inhibits their release of pro-inflammatory cytokines including granulocyte-macrophage colony-stimulating factor /GM-CSF, granulocyte colony-stimulating factor/G-CSF, IL-1 alpha, IL-1 beta, IL-6, IL-8 and TNF-alpha. Also interferes with antigen presentation by reducing the expression of MHC-class II and co-stimulatory molecules, thereby inhibiting their ability to induce T cell activation. In addition, controls the inflammatory response of macrophages by reprogramming essential metabolic pathways including mTOR signaling. |

Frequently Asked Questions
Common questions about Rat IL-10 (IL-10) Antibody Pair (STJAP00751)
This antibody pair is designed to detect rat Interleukin-10 (IL-10), corresponding to UniProt entry P29456. Reactivity has been confirmed for rat samples only.
Both antibodies are rabbit monoclonal IgGs supplied unconjugated at 1 mg/mL in 0.01M PBS, pH 7.2. Clone IDs are R07-7Q9 and R07-3Q4, and both have been affinity purified.
This antibody pair is validated for use in ELISA. No other assay formats are listed for this product.
Both antibodies were raised against a recombinant rat IL-10 protein. No specific immunogen region is defined in the product documentation.
Both components should be stored at 4°C for short-term use. For long-term storage, aliquot and keep at -20°C, avoiding repeated freeze/thaw cycles.
Working concentrations for both the capture and detection antibodies are to be determined by the end user. Titration experiments are recommended to establish optimal conditions for each specific assay setup.
