Additional Info
| Function | Major immune regulatory cytokine that acts on many cells of the immune system where it has profound anti-inflammatory functions, limiting excessive tissue disruption caused by inflammation. Mechanistically, IL10 binds to its heterotetrameric receptor comprising IL10RA and IL10RB leading to JAK1 and STAT2-mediated phosphorylation of STAT3. In turn, STAT3 translocates to the nucleus where it drives expression of anti-inflammatory mediators. Targets antigen-presenting cells (APCs) such as macrophages and monocytes and inhibits their release of pro-inflammatory cytokines including granulocyte-macrophage colony-stimulating factor /GM-CSF, granulocyte colony-stimulating factor/G-CSF, IL-1 alpha, IL-1 beta, IL-6, IL-8 and TNF-alpha. Also interferes with antigen presentation by reducing the expression of MHC-class II and co-stimulatory molecules, thereby inhibiting their ability to induce T cell activation. In addition, controls the inflammatory response of macrophages by reprogramming essential metabolic pathways including mTOR signaling. |

Frequently Asked Questions
Common questions about Mouse IL-10 (IL-10) Antibody Pair (STJAP00738)
This antibody pair is designed to detect mouse IL-10 (Interleukin-10), with reactivity confirmed for mouse. The target protein corresponds to UniProt entry P18893.
This antibody pair has been validated for use in ELISA. It is intended for research use only and is not validated for diagnostic or therapeutic applications.
Both the capture and detection antibodies are rabbit monoclonal IgGs supplied unconjugated at 1 mg/mL, formulated in 0.01M PBS at pH 7.2. Both are affinity purified, with clone IDs R01-1G2 and R04-3B4 respectively.
Optimal working concentrations are determined by the end user and are not predefined in the product specifications. Titration experiments are recommended to establish the appropriate dilution for each antibody in the intended assay format.
Both antibodies should be stored at 4°C for short-term use. For long-term storage, aliquots should be kept at -20°C, and repeated freeze/thaw cycles should be avoided to maintain antibody integrity.
Both antibodies in this pair were raised against a recombinant protein immunogen corresponding to mouse IL-10. The host species for both is rabbit, and both are monoclonal antibodies.
