Background Protein A is a cell wall protein ( ~ 56kDa ) isolated from Staphylococcus aureus. It consists of three regions : S is a processing signal sequence during secretion ; five homologous IgG binding domains E, D, A, B and C, as well as a cell wall anchoring region XM. The molecular weight of the truncated protein lacking the X region is about 31KD. Protein A can specifically bind to an amino acid sequence at the junction of heavy chain CH2 and CH3 in the Fc fragment of IgG, and can also bind to the heavy chain CDR2 and CDR3 regions in the Fab fragment. It has been demonstrated that the interaction between Protein A and IgG only involves the Fc region, and does not affect the binding of the Fab fragment to the antigen. Protein A binds to various human, mouse and rat IgG subclasses ( e.g. human IgG1, IgG2, IgG4 ; mouse IgG2a, IgG2b, IgG3 ; rat IgG2c ).
