Introduction:

CAMP responsive element binding protein 1 is synthesised by the CREB1 gene which helps with transcription activation, adipose cell differentiation and circadian rhythm synchronisation. Apart from its key functions in the human body, mutations of this gene lead to melanoma of soft tissues, several types of respiratory diseases and Coffin-Lowry syndrome. Furthermore, it can be used as a prognostic marker in favourable liver cancer. What is CREB1?

This gene encodes a transcription factor that is a member of the leucine zipper family of DNA binding proteins. This protein binds as a homodimer to the cAMP-responsive element, an octameric palindrome, and is phosphorylated by several protein kinases, and induces transcription of genes in response to hormonal stimulation of the cAMP pathway. Transcription activation is enhanced by the TORC coactivators which act independently of Ser-133 phosphorylation. Furthermore, this protein is involved in different cellular processes including the synchronization of circadian rhythmicity and the differentiation of adipose cells.

Where is CREB1 found?

CREB1 is mostly found in the nucleoplasm and on tissue level, it is evenly expressed throughout the majority of the tissues and has low brain region and immune cell specificity.

What role does CREB1 play in disease?

A CREB1 mutation has been found in a patient with multiple congenital anomalies consisting of agenesis of the corpus callosum, cerebellar hypoplasia, severe neonatal respiratory distress refractory to surfactant, thymus hypoplasia, and thyroid follicular hypoplasia.
