Introduction:

Chemerin Chemokine-Like Receptor 1 is a protein synthesised by the CMKLR1 gene and is responsible for inducing activation of intracellular signalling molecules and reduction of immune responses, enhancing adipogenesis and angiogenesis. It is mainly found in the lymphoid tissue and is immune cell enhanced, localised in plasmacytoid DC and intermediate monocyte. Several cancers and immune system disorders have been related to mutations or overexpressions of this gene, one of which is Monckeberg Arteriosclerosis. What is CMKLR1?

CMKLR1 is a protein coding gene responsible for synthesising the Chemerin Chemokine-Like Receptor 1 protein. This protein is a receptor for the chemoattractant adipokine chemerin/RARRES2 and for the omega-3 fatty acid derived molecule resolvin E1. Its interaction with RARRES2 induces activation of intracellular signaling molecules, such as SKY, MAPK1/3 (ERK1/2), MAPK14/P38MAPK and PI3K leading to multifunctional effects, like, reduction of immune responses, enhancing of adipogenesis and angionesis. Resolvin E1 down-regulates cytokine production in macrophages by reducing the activation of MAPK1/3 (ERK1/2) and NF-kappa-B. Positively regulates adipogenesis and adipocyte metabolism. This protein also acts as a coreceptor for several SIV strains (SIVMAC316, SIVMAC239, SIVMACL7E-FR and SIVSM62A), as well as a primary HIV-1 strain 

Where is CMKLR1 found?

CMKLR1 can be found mainly in the cell membrane, and is highly expressed in plasmacytoid DC and intermediate monocyte cells. It has cytoplasmic expression in most tissues, with higher expression in parathyroid gland and placenta.

What role does CMKLR1 play in disease?

Several diseases including cancer have been related with mutations of CMKLR1. One of them is Monckeberg Arteriosclerosis, presenting vessel hardening, consisting of calcium deposits in the muscle middle layer of arteries walls. Furthermore, CMKLR1 influences adipose tissue development, inflammation, and glucose homeostasis and may contribute to the metabolic derangement characteristic of obesity and obesity-related diseases.
