What is CASP3?
Caspase-3, also known as an executioner caspase in apoptosis, receives this name because of its role in the logistics of the destruction of cellular structures such as DNA fragmentation and degradation of cytoskeletal proteins. It is a heterotetramer which is made up of of two anti-parallel arranged heterodimers.
Where is CASP3 found?
Active Caspase-3 is mainly found in the cytosol and is highly expressed in the lungs, spleen, heart, liver and kidneys, and particularly in cells of the immune system. It is also found in lower levels the in brain and skeletal muscle, and rarely in testis.
What role does CASP3 play in disease?
CASP3 is a member of a family of Caspases. It is known that this particular member is involved in the neuronal death in Alzheimer's disease and cleavage of amyloid-beta 4A precursor. It is mainly involved in the activation cascade of caspases who take part in apoptosis execution. At the onset of apoptosis it proteolytically cleaves poly- (ADP-ribose) polymerase (PARP) and splits and activates SREBPs between the basic helix-loop-helix leucine zipper domain and the membrane attachment domain. This in turn activates caspase-6, -7 and -9 in downstream processes and takes part in the cleavage of huntingtin.
