• Western blot analysis of extracts of normal 293T cells and 293T transfected with Myc-NLK protein, using Myc-Tag antibody (STJ11103700) at 1:5000 dilution. Secondary antibody: HRP Goat Anti-mouse IgG (H+L) at 1:10000 dilution. Lysates/proteins: 25ug per lane. Blocking buffer: 3% non-fat dry milk in TBST. Detection: ECL Basic Kit. Exposure time: 1s.
  • Immunoprecipitation of NLK-Myc from 300 µg extracts of 293F cells transfected with a NLK expression vector containing a single C-terminal Myc-Tag was performed using 3 µg of Mouse anti Myc-Tag monoclonal antibody (STJ11103700). Mouse IgG isotype control was used to precipitate the Control IgG sample. IP samples were eluted with 1X Laemmli Buffer. The Input lane represents 10 % of the total input. Western blot analysis of immunoprecipitates was conducted using Mouse anti Myc-Tag monoclonal antibody at a dilution of 1:3000.
  • Immunofluorescence analysis of 293T cells transfected with Myc-Tag fusion protein and untreated 293T cells use mouse anti Myc-Tag monoclonal antibody (STJ11103700) at dilution of 1:100 (40x lens). Blue: DAPI for nuclear staining.

Background The protein encoded by this gene is a component of the exocyst complex, a multi-protein complex essential for the polarized targeting of exocytic vesicles to specific docking sites on the plasma membrane. Though best characterized in yeast, the component proteins and the functions of the exocyst complex have been demonstrated to be highly conserved in higher eukaryotes. At least eight components of the exocyst complex, including this protein, are found to interact with the actin cytoskeletal remodeling and vesicle transport machinery. This interaction has been shown to mediate filopodia formation in fibroblasts. This protein has been shown to interact with the Ral subfamily of GTPases and thereby mediate exocytosis by tethering vesicles to the plasma membrane. Alternative splicing results in multiple transcript variants.
