Introduction:

The conserved helix-loop-helix ubiquitous kinase is a protein synthesised by the CHUK gene and is part of the Cytokine-activated protein complex which is an inhibitor of the transcription factor NF-kappa-B complex. What is CHUK?

This gene encodes a member of the serine/threonine protein kinase family, which is a component of a cytokine-activated protein complex that is an inhibitor of the essential transcription factor NF-kappa-B complex and phosphorylates sites that trigger the degradation of the inhibitor via the ubiquination pathway, thereby activating the transcription factor. This protein is a serine kinase that plays an essential role in the NF-kappa-B signaling pathway which is activated by multiple stimuli such as inflammatory cytokines, bacterial or viral products, DNA damages or other cellular stresses. It acts as part of the canonical IKK complex in the conventional pathway of NF-kappa-B activation and phosphorylates inhibitors of NF-kappa-B on serine residues. These modifications allow polyubiquitination of the inhibitors and subsequent degradation by the proteasome. In turn, free NF-kappa-B is translocated into the nucleus and activates the transcription of hundreds of genes involved in immune response, growth control, or protection against apoptosis. It also phosphorylates RIPK1 at 'Ser-25' which represses its kinase activity and consequently prevents TNF-mediated RIPK1-dependent cell death 

Where is CHUK found?

CHUK is mainly localised in the nucleoplasm, cytosol and vesicles. It usually shuttles between the cytoplasm and the nucleus and on tissue level CHUK has cytoplasmic expression in most cell types and low tissue specificity.

What role does CHUK play in disease?

CHUK can be used as a prognostic marker in favourable renal cancer, and mutations can cause cocoon syndrome, characterised with multiple fetal malformations mainly visible on the face and seemingly absent limbs, because CHUK has an essential role in the development of skin epidermis.
