Frozen vs paraffin: choosing the right IHC antibody

Your tissue preparation method affects antibody compatibility. We stock IHC antibodies validated for both FFPE and frozen sections, ensuring reproducible results.

Formalin-fixed paraffin-embedded (FFPE) sections

IHC antibodies detect specific proteins in tissue sections using direct or indirect labelling strategies:

  • Primary antibodies (unconjugated or conjugated) bind directly to the target.
  • Secondary antibodies bind to the primary and amplify the detection signal.

    Direct staining: When multiplexing or working with low-background tissues, use conjugated primary antibodies (Alexa Fluor 488, Alexa Fluor 546, Alexa Fluor 647 , FITC, PE (Phycoerythrin), APC. Tri-color formats are also available for enabling multi-colour fluorescent imaging).

    Indirect staining: Utilise unconjugated primary antibodies with a matched immunohistochemistry secondary antibody to enhance sensitivity and signal amplification.

    For direct chromogenic IHC, primary antibodies conjugated to Horseradish Peroxidase (HRP) can be used together with common substrates, including 3,3'-diaminobenzidine (DAB) and Aminoethyl carbazole (AEC) to produce a visible colored precipitate for the localisation of target proteins.
